Predicting and controlling aggregation and fusion of plant protoplasts

The investigations presented in this thesis deal with heterokaryon formation of plant protoplasts-by means of electrofusion and with selection procedures
for the isolation of hybrid cells and calli. The purpose of the investigations was (I) to develop a model for protoplast electrofusion and use the model for the evaluation of external factors which influence heterokaryon yields, such as srrength of the electric field applied and conditions of protoplast
aggregation; (II) to design a universal technique for vital labeling of plant protoplasts for manipulation of the aggregation process and for identification and selection of heterokaryons. The fusion model and labelling procedure
were used in some practical experiments on regulatory effects heterokaryons and selection of hybrids.
