Controlled permeabilization of Escherichia coli outer membrane

In this thesis attention is focused on the permeabilization of the outer membrane of Escherichia coli. In chapter 2 a new procedure is presented which enables the transfer of macromolecules (e.g. DNA, lysozyme)a cross the outer membrane of Escherichia coli.. The method
is based on a Ca2+-competence procedure, EDTA treatment and a heat shock. The EDTA concentration used was especially crucial in faciltating outer membrane permeabilization; the necessary EDTA
concentration was a function of the cell density in the
permeabilization procedure. ...
